Product introduction:
Giemsa dye solution is composed of azure and eosin. Various cellular components have different chemical properties and different affinities for various dyes. The eosinophilic granule is a basic protein, which is combined with the acid dye eosin to be dyed pink; the basophilic particles such as nucleoprotein or lymphocyte cytoplasm are acidic substances, combined with the basic dye blue or azure, dyed It is purple-blue; the neutral particles are in an isoelectric state and can be combined with eosin and methylene blue, showing lavender. The various ingredients are distinguished by their different characteristics in combination with different substances in the Giemsa dye solution.
The Giemsa staining solution prepared by our company is prepared by using imported Giemsa dye as raw material, which can dye the nuclei into purple or blue-violet, dye the cytoplasm into pink, and show clear cells under light microscope. Dye the image. It is mainly used to show the difference in the shape and size of various blood cells in blood smears, with good dyeing effect, strong staining power and clear coloration.
Dyeing step: (for reference only)
Take 1ml of Giemsa Concentrate, mix well with 9ml of Giemsa Diluent, and use it.
1. Prepare the blood smear according to the conventional method. After the blood film is dry, fix it with methanol for 2 to 3 minutes. The prepared blood smear should have a suitable thickness and a uniform distribution so as not to affect the dyeing result.
2. Place the blood smear or bone marrow smear on the staining rack, add the diluted staining solution to cover the whole blood film, and stain for 15 to 30 minutes at room temperature.
3. Rinse slowly from the end of the slide with tap water (be careful not to pour the dye solution first or rinse the blood film directly), and then dry and then microscopically.
Precautions:
1. For your safety and health, wear a lab coat and disposable gloves.
2. If the staining is too dark or too light, please adjust the dyeing time or Giemsa working fluid concentration.
3. The Giemsa stock solution is prepared by conventional methods and filtered with filter paper. Please do not touch water when using. Otherwise, the time will be invalid.
4. PH has an effect on cell staining. The stained slides must be clean and free of acid and alkali contamination to avoid affecting the staining results.
