- Male breast cancer (MBC) is a very rare type of cancer, accounting for only 1% of all breast cancer cases. Currently, the pathogenic genes and treatment methods for MBC are still poorly understood. In an article published in the March issue of the international academic journal Cold Spring Harbor Molecular Case Studies, researchers from Italy and the United States found a new genetic variant in hormone receptor-positive (HR+) MBC patients, One study found differences in genetic variation in 10 MBC patients.
The author of the article described the treatment history of the HR+ MBC patient. By using the PI3K/mTOR inhibitor drug BEZ235 to target the PI3K/mTOR signaling pathway, combined with everolimus as a third-line treatment, the patient's metastatic ductal adenocarcinoma was stabilized and had a long period of stability. But after 18 months, he began to resist the treatment and his condition was further developed. Researchers used genomics and immunohistochemical analysis to analyze why patients benefit from this combination therapy and why they are resistant to the therapy.
The researchers performed a full exon sequencing of the tissue samples before treatment and further progression of the disease, and used whole blood as a normal control. It was found that a region of chromosome 12p of the patient was deleted in the resistant tumor, and the expression of HR protein in the resistant tumor cells also increased.
This study provides insights into the drug response of male breast cancer and patients to the combination of BEZ235/everolimus, and also deepens our understanding of the development and resistance of MBC disease.
