The reporter learned from the Hefei Institute of Material Science of the Chinese Academy of Sciences that Zhang Zhongping, a researcher at the Institute of Intelligent Micro-Nano Devices, has made a breakthrough in tumor cell detection and precision surgery navigation. Relevant research results have been published in the international chemical journal "ACS Nano" and have applied for national invention patents.

The uncontrolled growth and immortalization of tumor cells requires the catalysis of highly active telomerase. The researchers used gold nanoparticles as a carrier to design a spherical nucleic acid probe with a large amount of specific double-stranded DNA on the surface. Under the catalysis of telomerase, the probe can release fluorescent dye into the cytoplasm, causing the tumor cells to emit red fluorescence, thereby achieving visual detection of tumor cells. Through the change of fluorescence signal, the precise differentiation of more than a dozen representative tumor cells from normal cells, the identification of the malignant degree of tumor cells, and the in vivo imaging, naked eye visualization and tissue section identification of mouse tumors were realized. The results show that this spherical nucleic acid probe is an ideal tumor precision surgical navigation contrast agent.

The current tumor surgery is mainly based on the doctor's experience and subjective judgment, and lacks effective means to distinguish tumor tissue from normal tissue. This study allows tumor cells to "glow" through spherical nucleic acid contrast agents. It can help doctors to determine the location and boundary of tumors at the cellular level by virtue of their own positioning accuracy and high sensitivity, so as to accurately remove the lesions and reduce the risk of surgery. At the same time, the recurrence rate and mortality after tumor surgery are greatly reduced. It is reported that the team will further carry out animal and clinical trials of precise tumor resection, and finally develop contrast agents for clinical tumor precision surgery and promote industrial application.
